About the origins of enhanced visualization of amyloid plaques on APP/PS1 mouse brain using GdDOTA passive staining

چکیده
Introduction T1-relaxating contrast agent (e.g. GdDOTA) “passive staining” is a smart strategy to enhance signal-to-noise ratio (SNR) for MRI microscopy (μMRI) of ex vivo tissue samples. The principle of the method is to immerse organs (e.g. brains) in a solution of GdDOTA and let it diffuse for days in order to reduce the T1 and further reduce the optimum relaxation time (TR) and accordingly the acquisition time. Such strategy has been adopted successfully to detect amyloid plaques in high-resolution T2 -weighted images of APP/PS1 transgenic mice brain [1]. Nevertheless, mechanisms of this contrast enhancement of amyloid plaques were not thoughtfully investigated. In the present study, we have carried such investigation and established how GdDOTA is diffusing throuh tissues during passive staining and what the consequences are for the contrast of amyloid plaques vs. healthy brain tissue.

Cortical iron regulation and inflammatory response in Alzheimer's disease and APPSWE/PS1ΔE9 mice: a histological perspective
Disruption of iron homeostasis and increased glial response are known to occur in brains afflicted by Alzheimer's disease (AD). While the APP/PS1 transgenic mouse model recapitulates the hallmark amyloid-beta plaque pathology of AD, it does so in a different neuronal mileu than humans. Understanding the iron characteristics and glial response of the APP/PS1 model is important when testing new t...
متن کاملEarly induction of oxidative stress in a mouse model of Alzheimer’s disease with heme oxygenase activity
Evidence suggests that brain tissues of patients with Alzheimer's disease (AD) are easily attacked by oxidative stress, and numerous studies indicate that heme oxygenase (HO) is a major cell adaptive responder to stress. However, whether HO‑1 and HO‑2 play different roles in this process has not yet been studied. In the present study, it was shown in an AD model that HO‑1 and HO‑2 have differen...
متن کاملAutomatic detection of amyloid plaques on ex vivo APP/PS1 mouse brain using a zoom T2-weighted spin echo sequence
Introduction Amyloid plaques are known as a marker of Alzheimer's disease. They are traditionally detected as hypointense spots on T2-weighted images taking advantage of the susceptibility effect due to iron embedded inside them. However it has been recently reported that T2 contrast might be more efficient to detect cortical amyloid plaques [1]. In this context, the goal of the present study w...
متن کاملGalactic Cosmic Radiation Leads to Cognitive Impairment and Increased Aβ Plaque Accumulation in a Mouse Model of Alzheimer’s Disease
Galactic Cosmic Radiation consisting of high-energy, high-charged (HZE) particles poses a significant threat to future astronauts in deep space. Aside from cancer, concerns have been raised about late degenerative risks, including effects on the brain. In this study we examined the effects of (56)Fe particle irradiation in an APP/PS1 mouse model of Alzheimer's disease (AD). We demonstrated 6 mo...
متن کاملAging increased amyloid peptide and caused amyloid plaques in brain of old APP/V717I transgenic mice by a different mechanism than mutant presenilin1.
Aging of transgenic mice that overexpress the London mutant of amyloid precursor protein (APP/V717I) (Moechars et al., 1999a) was now demonstrated not to affect the normalized levels of alpha- or beta-cleaved secreted APP nor of the beta-C-terminal stubs. This indicated that aging did not markedly disturb either alpha- or beta-secretase cleavage of APP and failed to explain the origin of the ma...
